Konsument:

Betäubungsmittel (opioid -) Analgetikum Agenten sind extensiv metabolisiert durch die Leber, und einige von Ihnen (zB, Codein, hydrocodone, meperidin, Methadon, Morphin, propoxyphene) haben aktive Metaboliten, die weiter umgewandelt, um inaktive Substanzen. Die serum-Konzentrationen dieser Wirkstoffe und deren Metaboliten erhöht und die Halbwertszeiten verlängert bei Patienten mit eingeschränkter Leberfunktion. Therapie mit Opioiden verabreicht werden soll vorsichtig und initiiert bei reduzierten Dosierungen bei Patienten mit Lebererkrankungen. Die nachfolgenden Dosen sollten titriert werden, basierend auf der individuellen Reaktion eher als eine Feste Dosierung.

Phenothiazine sind umfassend durch die Leber metabolisiert und können sich bei Patienten mit Leberinsuffizienz. Darüber hinaus ist die Verwendung von einige Phenothiazine wurden im Zusammenhang mit negativen hepatische Effekte einschließlich cholestatische Gelbsucht und erhöhte Leberenzyme, in der Regel innerhalb der ersten paar Monate der Therapie. Cholestatische Gelbsucht tritt in der Regel zwischen der zweiten und vierten Woche der Therapie in etwa 0,1% bis 4% aller Patienten. Therapie mit Phenothiazine gegeben werden sollte vorsichtig in Patienten mit bereits bestehenden Leber-Krankheit, Leber-Enzym-Anomalien, oder hepatitis. Die Funktion der Leber und Urin-bilirubin-tests sollten durchgeführt werden, in regelmäßigen Abständen während einer längeren Therapie, und die Patienten sollten angewiesen werden, sofort über jedes Anzeichen oder Symptome andeutend cholestatische Gelbsucht, wie Schmerzen im Oberbauch, übelkeit, Gelbfärbung der Haut, grippeähnliche Symptome, Hautausschlag und Fieber. Phenothiazin-Therapie sollte abgebrochen werden, vorzugsweise dauerhaft, wenn eine Gelbsucht Auftritt, und ist auf die Droge. Klinische recovery ist in der Regel beobachtet, die innerhalb von ein paar Wochen nach Entzug von Therapie, obwohl histopathologische Veränderungen fortbestehen können für längere Zeiträume.
